N-Methyl-4-pyridone-5-carboxamide as a metabolite of nicotinic acid in man and monkey.

In previous experiments reported from this laboratory (1) N-methyl-4-pyridone-5-carboxamide was isolated from rat urine and identified as the major urinary metabolite of the pyridine nucleotides. Recently Wieland et al. (2) synthesized this 4-pyridone and have found their synthetic product to be identical with the compound isolated from rat urine. Experiments have now been carried out to determ...

The excretion of N-methyl-2-pyridone-5-carboxamide by man following ingestion of several known or potential precursors.

The development of a rapid method for the determination of N-methyl2-pyridone-5-carboxamide (pyridone) in urine (1) has made it practical to attempt to confirm and extend the studies of Holman and de Lange (2) and others (3-5) concerning the excretion of the pyridone following the administration of various known or possible precursors to man. In the conversion of nicotinic acid to the pyridone ...

Channel-forming solvate crystals and isostructural solvent-free powder of 5-hydroxy-6-methyl-2-pyridone.

Crystals of 5-hydroxy-6-methyl-2-pyridone, (I), grown from a variety of solvents, are invariably trigonal (space group R3); these are 5-hydroxy-6-methyl-2-pyridone acetone 0.1667-solvate, C(6)H(7)NO(2)·0.1667C(3)H(6)O, (Ia), and 6-methyl-5-hydroxy-2-pyridone propan-2-ol 0.1667-solvate, C(6)H(7)NO(2)·0.1667C(3)H(8)O, (Ib), and the forms from methanol, (Ic), water, (Id), benzonitrile, (Ie), and b...

N-methyl-2-pyridone-5-carboxamide (2PY)—Major Metabolite of Nicotinamide: An Update on an Old Uremic Toxin

N-methyl-2-pyridone-5-carboxamide (2PY, a major metabolite of nicotinamide, NAM) was recently identified as a uremic toxin. Recent interventional trials using NAM to treat high levels of phosphorus in end-stage renal disease have highlighted new potential uremic toxicities of 2PY. In the context of uremia, the accumulation of 2PY could be harmful-perhaps by inhibiting poly (ADP-ribose) polymera...

Effects of 1-methylnicotinamide and its metabolite N-methyl-2-pyridone-5-carboxamide on streptozotocin-induced toxicity in murine insulinoma MIN6 cell line.

1-methylnicotinamide (MNA) is a primary metabolite of nicotinamide. In recent years several activities of MNA have been described, such as anti-inflammatory activity in skin diseases, induction of prostacyclin synthesis via COX-2, aortal endothelium protection in diabetes and hypertriglyceridaemia and increased survival rate of diabetic rats. 1-methylnicotinamide was also suggested to protect p...

Purification and properties of the aldehyde oxidases from hog and rabbit livers.

The aldehyde oxidases from rabbit and hog livers have been purified to homogeneity by modifications of and additions to previously published procedures. The final preparations were judged to be homogeneous on the basis of ultracentrifugal, electrophoretic, chromatographic, spectral, and kinetic criteria. A number of physical and kinetic properties of the homogeneous enzymes have been determined...
